盐胁迫下西伯利亚蓼蛋白质双向电泳分析及质谱鉴定  被引量:4

摘  要:通过蛋白质双向电泳和质谱技术,对盐胁迫前后西伯利亚蓼的蛋白质组进行了比较分析.在pH 3~10范围内,在西伯利亚蓼地下茎蛋白表达图谱中发现23个蛋白质点胁迫后的表达量与对照有明显差异(±2倍以上),其中包括一个新增蛋白点.对表达量变化较大的11个蛋白点进行质谱分析,并通过数据库检索进行蛋白质鉴定与功能预测,确定了4种蛋白质.其中的两种蛋白甘油醛-3-磷酸脱氢酶(Glyceraldehyde 3-phosphate dehydrogenase)和丙酮酸正磷酸二激酶(Pyruvate,orthophosphate dikinase)胁迫后高表达,说明地下茎细胞内正进行着旺盛的呼吸代谢,以提供维持植物生长发育以及抵抗外界胁迫所需的能量.Proteins in Polygumn sibiricum under salt stress and non-stress were compared and analyzed by two-dimensional gel electrophoresis(2-DE) and identified by mass spectrometry.In the range of pH为3~10,the expression levels of 23 proteins have obvious differences(±more than 2 fold) in proteomic map of rhizome from Polygumn sibiricum between stress and non-stress group, including a new protein spot.11 proteins expressed in high level were identified using matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS),and then search in the data base for protein identification and function prediction.Four proteins were confirmed,two proteins in which named Glyceraldehyde 3-phosphate dehydrogenase and Pyruvate,orthophosphate dikinase expressed a high level after stress.This indicated that bloom respiration metabolism was doing in the rhizomatous cell to support energy for plant growth and resist stress.
