摘  要:目的观察17β雌激素对β淀粉样蛋白25~35片段(Aβ25~35)所致PC12细胞毒活性影响的可能机制。方法用逆转录一聚合酶链反应(RT PCR)技术,观察雌激素对Aβ25~35损伤的PC12细胞的Bcl^2mRNA及Bax mRNA表达的影响。结果用Aβ25~35处理PC12细胞24h,与对照组相比,Bcl^2mRNA的表达水平降低,Bax mRNA的表达水平升高;而Aβ25~35加17β雌激素引起Bcl^2mRNA的表达水平较单纯Aβ处理组增高,Bax mRNA的表达水平较单纯Aβ处理组降低。结论17β雌激素在Aβ25~35所致PC12细胞毒性的神经保护作用中,其机制是通过上调Bcl^2mRNA的表达与下调Bax mRNA的表达来实现的。Objetive To study the potential neuroprotective mechanisms of 17β-estradiol on Aβ_ 25~35 -induced cytotoxicity in PC12 cells.Methods PC12 cells were exposed to Aβ_ 25~35 or treated with 17β-estradiol before exposure to Aβ_ 25~35 ,the expression of Bcl-2 mRNA and Bax mRNA was measured by reverse transcription polymerase chain reaction (RT-PCR).Results Compared with control group, the expression of Bax mRNA was increased and the expression of Bcl-2 mRNA was decreased in the PC12 cells which treated with Aβ_ 25~35 for 24 hours. And compared with Ap group, the expression of Bcl-2 mRNA was increased and the expression of Bax mRNA was decreased in the PC12 cells which treated with 17β-estradiol before exposure to Aβ_25~35 . Conclusions The neuroprotection of 17β-estradiol in Aβ_25~35 -induced cytotoxicity of PC12 cells was via up-regulation of Bcl-2 mRNA and down-regulation of Bax mRNA.
