盐酸多奈哌齐、西酞普兰对局灶性脑缺血再灌注大鼠认知功能的影响

摘  要:目的观察盐酸多奈哌齐、西酞普兰对局灶性脑缺血再灌注大鼠认知功能的影响,方法将36只SD大鼠雌雄各半随机分为假手术组(A组),缺血90 min再灌注0.9%氯化钠溶液治疗组(B组)、盐酸奈哌齐治疗组(C组)和西酞普兰治疗组(D组)。采用Zea Longa's线栓法制备SD大鼠大脑中动脉闭塞(MCAO)模型,评定术后B、C、D组大鼠神经行为学评分,观察A组、B组术后14 d红四氮唑(TTC)染色以及各组水迷宫实验及听觉事件相关电位(P300)测试。结果术后大鼠神经行为学评分均>2分。B组大鼠术后第14 d TTC染色的结果显示了模型的可靠性。定位航行试验:实验的第2~5 d,B组大鼠逃避潜伏期显著长于A组(P<0.01);C组、D组大鼠逃避潜伏期显著短于B组(P<0.01);实验的第3~5 d,C组大鼠逃避潜伏期显著短于D组(P<0.05)。空间探索试验:C组、D组大鼠平台象限游泳时间和总游泳时间的比值(tP/tT)、平台象限距离和总距离的比值(dP/dT)均显著大于B组(P<0.05);B组大鼠的dP/dT、tP/tT值均显著小于A组(P<0.01);C组dP/dT、tP/tT值均显著大于D组(P<0.05)。4组大鼠听觉事件相关电位间差别有显著性意义(P<0.01)。结论大鼠大脑中动脉闭塞90 min后再灌注能导致认知功能下降,盐酸多奈哌齐和西酞普兰均能改善大鼠认知障碍,前者的效果可能更好。Objective To observe the variation of cognition ability of the middle cerebral artery occlusion (MCAO) rats following cerebral ischemia-repcrfusion and the effect of Donepezil and Cipramil on cognition function.Methods Thirty- six SD rats were randomly divided into the sham group (group A,n=12) ,the ischemia 90 min and reperfusion with Saline treatment group (group B,n=12) ,Donepezil group (group C,n=6) and Cipramil group (group D,n=6).The models of MCAO were made by occluding the MCA with thread according to Longa's method.Behavioral assessments were evaluated by neurological test.Brain tissue damage was detected 14d after the surgery by triphenyl tetrazolium chloride (TFC) staining.Cog- nition ability was determined by Morris-maze test and P300 test.Results The scores of neurological test of ischemia groups were all above 2 points.The TFC staining results of infarct areas of group B ascertained the reliability of Longa's method.After the first day in place navigation test,escape latency (EL) of group B was significantly longer than that of group A (P<0.01); while EL of group C and D was significantly shorter than group B(P<0.01).EL of group C was significantly shorter than group D after the third day (P<0.05).The ratio of dP/dT and tP/tT of group C was significantly higher than group B and D in spatial probe test.The ratio of dP/dT and tP/tT of group A was significantly higher than group B.Significant differences were found between each group in P300 test.Conclusion The ischemia 90 rain and reperfusion induced by MCAO can result in cog- nitive impairment.Donepezil and Cipramil are shown to improve significantly the cognitive function and the former may be more efficient.
