普洛迪对局灶性脑缺血大鼠的神经保护作用研究

摘  要:目的研究普洛迪对局灶性脑缺血大鼠的神经修复作用。方法采用Longa法制备SD大鼠大脑中动脉缺血模型(MCAO),分为:假手术组;持续缺血组;药物干预(普洛迪、金纳多、施普善)组。应用免疫组化法观察巢蛋白(Nestin)和神经元特异性烯醇化酶(NSE)的表达变化,并使用多媒体彩色病理图像分析系统进行定量分析;利用草酸-焦锑酸钾电镜细胞化学技术观察缺血脑组织的超微结构改变。结果药物干预组的病变明显轻于持续缺血组;其中,普洛迪(高剂量)组优于金纳多、施普善组。普洛迪(高剂量)组Nestin的表达升高最显著,NSE的表达下降程度最轻,且呈显著的普洛迪剂量依赖性。结论普洛迪可使缺血后Nestin表达上调,并减轻NSE表达下降程度,从而起到减轻缺血性脑损伤的作用。Objective To study the neurological protective effects of Plaudit^(TM) after focal cerebral ischemia in rats. Methods Rats model of ischemia of cerebral middle artery occlusion (MCAO) was established according to Longa pattern.The rats were randomly divided into sham-operation group,persistent ischemia group and drugs treated groups including Plaudit^(TM), Ginaton^(?) and Cerebrolysin^(?).Using immunohistochemical staining method and color image analysis,we measured the expression of Nestin and Neurone Specific Enolase (NSE) in rats' brain tissues.Ultra-structure of brain tissues was observed by transmis- sion electron microscope.Results Pathological changes in drugs treated groups were obviously less than those in persistent is- chemia group.Pathological changes in high dose of Plaudit^(TM) treated subgroup were less than those in Ginaton^(?) or Cerebrolysin^(?) treated groups.In high dose of Plaudit^(TM) treated subgroup,extent of increased expression of Nestin was the most obvious and that of decreased expression of NSE was the lightest.Besides,expression of Nestin and NSE showed significant Plaudit^(TM) dose-de- pendent.Conclusion Plaudit^(TM) may produce protective effects by enhancing expression of Nestin and lessening extent of expres- sion of NSE after focal cerebral ischemia.
