摘  要:为了便于进行疟疾疫苗保护性观察及建立恶性疟原虫克隆株的需要,对虫库保存的恶性疟原虫FCC1/HN分离株进行某些生物特性鉴定。通过体外培养条件变更以观察现存FCC1/HN分离株恶性疟原虫的生长特性,以体外微量法测定该株原虫对氯喹的敏感性及用套式PCR/RFLP分析方法对该株原虫二氢叶酸还原酶基因型进行鉴定。结果显示,现存FCC1/HN株恶性疟原虫对体外培养条件适应性强,对氯喹高度敏感,DHFR基因的第108号编码为AAC突变型。建议对虫库保存的恶性疟原虫FCC1/HN分离株的生物活性定期进行鉴定。To use conveniently the FCC1/HN of Plasmodium falciparum in these field of verification of malaria vaccine; s protection and estiblisment of the cloned strain of Plasmodium falciparum , we deterninted it's biological activity. Observation of the growing character of the separate strain in vitro culture, its sensity to drug by microtest and DHFR genetype by nested PCR/ RFLP. The storing FCC1/HN strain of Plasmodium falciparum adapt powerfully to the condition under in vitro AAC sequence at the 108 codon of DHFR gene, which the 108 aminocide of DHFR is asparagine so that the strain hsve resistance to Pyrimethamine. It will be necessory to determine regularly the biological activity for reasonably usage the FCC1/HN strain of Plasmodium falciparum.
