摘 要:目的:探讨人参二醇组皂苷(Panaxadiolsaponins,PDS)对动脉粥样硬化大鼠血脂、血清NO、MDA含量、SOD活力的影响.方法:高脂饮食加维生素D3(总量7×105U/Kg,分别于实验第1周和第6周腹腔注射),建立大鼠动脉粥样硬化模型.选用健康Wistar大鼠45只,体重180~230g,雌雄不限.按清洁级动物分笼喂养,适应性喂养1周后随机分为3组:正常对照组,以普通饲料喂养;高脂模型组,以高脂饲料喂养;PDS给药组,高脂饲料喂养,同时每日1次PDS[100mg/(kg·d)]灌胃,前两组以相应生理盐水灌胃,每周称重1次,并根据体重变化调整给药量.11周后颈动脉采血,检测血脂水平,血清一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平.结果:1、PDS对血脂的影响:与对照组相比较,模型组大鼠血清TC、LDL-c及AI显著升高(P<0.01);与模型组比较,PDS给药组TC、LDL-c和AI明显降低,差异显著(分别P<0.01,P<0.01,P<0.05);而血清甘油三脂、高密度脂蛋白水平相比无统计学意义.2、PDS对血清NO含量的影响:与对照组相比较,模型组大鼠血清NO含量明显降低,P<0.01;与模型组比较,PDS组NO含量明显增高,P<0.05;药物组与对照组相比无统计学意义.3、PDS对血清MDA含量与SOD活力的影响:模型组MDA水平与对照组相比明显升高,P<0.01;而SOD活力明显降低,P<0.01.与模型组相比,PDS对血清SOD活力有明显升高的作用,P<0.05;同时,PDS具有显著降低MDA水平的作用,P<0.05.PDS组与对照组相比无统计学意义.结论:1、PDS通过降低大鼠血清总胆固醇、低密度脂蛋白水平,发挥其抗AS作用.2、PDS能够提升高脂大鼠血清NO水平,从而保护血管内皮,抑制AS的形成.3、PDS能够提高血清SOD活力,降低MDA含量,增强清除氧自由基和抗脂质过氧化功能,从而稳定细胞膜,保护内皮而发挥抗AS作用.Objective:To investigate the effects of Panaxadiol saponins on lipemia、NO 、MDA and SOD in the atherosclerosis Rats.Methods:Experimental atherosclerosis in rats was produced by feeding with high fat diet and administering vitamin D3 (7 × 10~5 U/Kg) in intraperitoneal injection at the first week and the sixth week respectively.Healthy forty-five male or female Wistar rats, whose body weights range from 180g to 230g,were randomized into 3 groups after being well adapted to a new life for a week.The three groups were normal group (feed on normal diet),AS model group (feed on high fat diet) and PDS group (feed on high fat diet and PDS 100mg/kg · d).The former two groups were given isotonic sodium chloride instead of PDS accordingly.The rats of each group were weighed once a week and the dosages were adjusted to the changes of body weight accordingly.After the rats were fed for successive 11 weeks,serum levels of total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C),Nitric oxide (NO),Superoxide dismutase (SOD),Malondialdehyde (MDA) were detected respectively.Results:1.The effect of PDS on blood lipoproteins:The serum level of TC 、LDL-c and AI of Model group was significantly higher than that of Control group(P <0.01);PDS decreased the serum level of TC,LDL-c,and AI significantly. The difference of each index between Model group and PDS group was remarkable (P < 0.01,P < 0.01,P < 0.05) but there was no sta- tistic difference between them in TG and HDL-c.2.The effect of PDS on the serum NO level:The serum NO level of PDS group was higher than that of model group (P < 0.05).The NO level of control group was higher than that of model group (P < 0.01);The difference in NO level between PDS group and control group was not remarkable statistically.3.The effects of PDS on the serum MDA level and SOD activity:MDA level of model group increased obviously compared with that of control group(P < 0.01).SOD activity of Model group decreased obviousl
