摘 要:目的识别和确定与广西巴马地区长寿人群相关的基因。方法采用长寿对照方法,分组:129名年龄(94~113)岁的长寿者为长寿组,长寿者家人年龄不大于 59岁的 179人为年龄对照组,当地非长寿自然人群153人作为自然人群对照组,进行ApoE基因分型,分型方法采用PCR-RFLP,并经测序确定。分别比较长寿组与各组人群的基因型、基因频率的差别、并进行显著性检验。各个对照组的ApoE基因频率均符合Hardg—Weinberg平衡。结果①长寿组与其亲属年龄对照的ApoE基因型和基因频率相比之间均无显著性差别,表明两者属同一人群,且年龄变化与基因频率无关。②长寿组与当地自然人群对照组相比较有显著性阳性关联(P=0.0001,P=0.04)的为ε2/ε2、ε3/ε3基因型。其中ε3/ε3纯合子是最主要的长寿关联基因。Objective To identify and reconfirm the gene associated with long- lived elderly in Red River area in Guangxi in China Methods Applying the case -control method, we classified the 129 longevity families containing the family members 436 individuals into different groups. They included: longevity group with 129 long-lived elderly probands, age range from 94 to 113 years old; age control group, 179 persons aged less than 60 years old from the longevity fami- lies; and local residents 153 persons age range 60~80 years old as control group. We developed lab genotyping by way of PCR-RFLP technique, and com- pared the differences of allele, genotype of longevity group with the other groups. Pesults 1. There were no significant differences of genotype and allele frequencies between longevity and its age matched 8 groups, it showed that both subjects belong to same genetic origin and no effect of age increment on gene fre- quencies. 2. There were significant positive association(P=0.001, P=0.04) for genotypes ε2/ε2 (5.34%; 0%) and ε3/ε3 (79.39%; 68.63%), through compa- ring longevity group with local control group. Conclusion we find out and con- firm the ApoE ε2/ε2 and ε3/ε3 genotypes associated with longevity in the popu- lation in Red River area in Guangxi in China,especially homozygote ε3/ε3 geno- type is perhaps the major gene associated with longevity.
