淫羊藿甙对小鼠同种异基因皮片移植的影响

摘　　要：目的:研究淫羊藿甙(ICA)对小鼠同种异基因皮片移植的影响。方法:试验分为腹腔注射生理盐水、ICA、环孢素A(CsA)以及CsA与ICA联用4组,5 d后将C57BL/C小鼠的背部皮片移植到BALB/c小鼠的相同位置,用眼科缝合线缝合,观察皮片愈合情况并记录皮片外观性状、排斥时间,于第11天取移植皮片做病理石蜡切片,取淋巴结细胞和脾细胞做增殖实验(CFDA-SE染色)。结果:CsA+ICA联用能够延长皮片排斥时间,使皮片愈合得更好,胸腺和脾脏指数均比单独CsA组高,切片显示浸润的炎症细胞也较少,其淋巴结细胞增殖能力明显降低。结论:CsA与ICA联合能抑制移植造成的免疫排斥反应,可能是通过抑制受体小鼠的反应性T细胞的活化增殖来降低细胞免疫反应。Aim:To study the effect of icariin(ICA) on the rejection time of skin and proliferation of murine thymus and spleen cells. Methods: There are four groups in the experiment: peritoneal injection of saline group,ICA group,Cyclosporine A(CsA) group and CsA accompanied with ICA group.Five days later,the back skin grafts of C57BL/C mice were transplanted onto the same place of BALB/c mice and sewed up with ophthalmological sutural line.Observe the concrescence of skins and record their external characters and rejection time.The skins were taken for making pathological paraffin section and lymphocytes and spleen cells were used for proliferation detection.Results: CsA+ICA can lengthen the rejection time and make the skin recover better with less infiltrative inflammatory cells.Its thymus and spleen index are both higher than CsA group's while its proliferation ability becomes lower.Conclusion:CsA and ICA may inhibit the immune rejection caused by transplantation through restraining the activation and proliferation of the reactive T cells of the recipient mouse.
