摘 要:目的:观察过氧化物酶体增殖物激活受体γ激动剂(GW1929)对小鼠腹腔巨噬细胞源泡沫细胞的形成及基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶-9(MMP-9)分泌的影响。方法:氧化型低密度脂蛋白(oxLDL)(50μg/mL),过氧化酶体增殖物激活受体γ激动剂1929(GW1929)(20μmol/L)作用于小鼠腹腔巨噬细胞24 h后,油红O染色观察各组泡沫细胞的形成。并用ELISA法测定各组培养液上清中MMP-2、MMP-9的浓度。结果:对照组及GW1929组细胞几乎未被油红O染色,oxLDL组大量细胞胞质被油红O染成暗红色,而oxLDL+GW1929组细胞胞质染色明显浅于oxLDL组细胞。oxLDL组MMP-9的浓度明显高于对照组及GW1929组(P<0.05)。而oxLDL+GW1929组MMP-9的浓度明显低于oxLDL组(P<0.05)。而各组间MMP-2的浓度并没有差别(P>0.05)。结论:GW1929可能具有稳定粥样斑块的作用,其机制可能与其抑制泡沫细胞的形成及MMP-9的生成有关。Aim:To observe the effect of peroxisome proliferator-activated receptor γ agonist,GW1929 on mouse peritoneal macrophage foam cell formation and matrix metalloproteinases-2(MMP-2)、MMP-9 secretion. Methods: The mice peritoneal macrophages were treated with oxLDL(50 μg/mL)and GW1929(20μmol/L)for 24 h.Then,they were stained by oil red O to observe foam cell formation.Further more,concentrations of MMP-2,MMP-9 in the media of macrophage were detected.Results: The mice peritoneal macrophages in controlled group and GW1929 group were not stained by oil red O,but the macrophages in oxLDL group become dark red after stained by oil red O,and color of macrophages in oxLDL+GW1929 group is shallower than that in oxLDL group.The concentration of MMP-9 in oxLDL(50 μg/mL)group was higher than that in control group and GW1929 group obviously(P<0.05),but the concentration of MMP-9 in oxLDL(50 μg/mL)+GW1929 group was lower than that in oxLDL(50 μg/mL)group(P<0.05).There was no difference in MMP-2 concentration among various groups(P>0.05).Conclusion:GW1929 probably stabilize atheromatous plaque,which may be related to inhibiting foam cell foamation and MMP-9 secretion.
