摘 要:目的:探讨三羟异黄酮(GEN)联合阿霉素(ADM)对人乳腺癌耐药细胞MCF-7/ADM中HER-2/neu mR-NA及蛋白表达的影响。方法:将不同浓度GEN和ADM单独或联合作用于体外培养的MCF-7/ADM细胞后,RT-PCR检测HER-2/neu mRNA表达情况,Western blot检测HER-2/neu蛋白表达情况。结果:体外培养的MCF-7/ADM细胞高表达HER-2/neu mRNA及蛋白,经GEN单独及联合ADM作用后,HER-2/neu mRNA和蛋白表达明显下调。联合用药比相同浓度GEN单用下调更明显。结论:人乳腺癌MCF-7/ADM细胞耐药性与HER-2/neu mRNA及蛋白高表达相关,GEN能部分逆转MCF-7/ADM细胞多药耐药性。Objective To investigate the effecs of genistein combined with adriamycin on the HER-2/neu mRNA and protein expression in human breast cancer MCF-7/ADM cells.Methods The cultured MCF-7/ADM cells were treated with different concentrations of genistein and adriamycin in vitro.The expression of HER-2/neu mRNA and protein were determined with RT-PCR and Western blot,respectively.Results The HER-2/neu mRNA and protein were highly expressed in cultured MCF-7/ADM cells in vitro,which were significantly decreased after treated with genistein single or combined adriamycin.The decreasing effect with combined administration were more obviously than with single genistein administration at the same concentration.Conclusion The drug resistance of human breast cancer MCF-7/ADM cells may be related to the high expression of HER-2/neu mRNA and proteins,Genistein could partly reverse the multidrug resistance of MCF-7/ADM cells.
