摘 要:目的:研究从味连新鲜植株中提取的DNA与干燥药材基因组DNA的差别。方法:在改良的CTAB法基础上对味连新鲜植株与干燥药材提取总DNA,采用紫外分光光度法及琼脂糖凝胶电泳法检测DNA纯度与浓度。结果:从新鲜植株中提取的DNA浓度较高,从干燥药材中提取的DNA有降解现象。结论:味连的新鲜植株及干燥药材中均可以提取DNA,但中药材的干燥、加工、贮藏等过程会对DNA有一定的降解。Objective:To research the differences of genomic DNA extraction between plant and dried medicinal materials.Method:The genomic DNA of Coptis chinensis was extracted by modified cetyltrimethyl ammonium bromide(CTAB)method,and the concentration and purity of DNA was detected by UV spectrophotometry and agarose gel electrophoresis method.Result:The DNA concentration of Coptis chinensis plant was higher than the dried rhizoma coptidi,and the dried rhizoma coptidi DNA was degraded.Conclusion:The genomic DNA of Coptis chinensis can be extracted from plant or dried rhizoma coptidi,but the rhizoma coptidi DNA can be degraded to some extent as the result of its' drying,processing and preservation procedures.
