摘　　要：目的:探讨Ⅱ型cGMP依赖性蛋白激酶(tpyeⅡcGMP dependent protein kinase,PKGⅡ)在原发性胃癌组织中的表达及其在胃癌发生发展中的意义。方法:选择50例原发性胃癌组织、50例正常胃黏膜组织,应用免疫组化SP法和逆转录PCR法分别检测PKGⅡ蛋白和mRNA表达水平。结果:PKGⅡ在正常胃黏膜组织中表达较高,在胃癌组织中表达明显降低(P<0.05),相关性分析结果显示PKGⅡ的表达与胃癌细胞的分化程度相关。结论:PKGⅡ在胃癌组织中表达明显降低,可能与胃癌的发生发展有关。Objective: To investigate the expression of typeⅡ cGMP dependent protein kinase(PKGⅡ) in human primary gastric cancer,and analyze its significance in the tumorigenesis and development of gastric cancer.Methods: Fifty cases of primary gastric cancer and 50 cases of normal gastric mucosa tissue were chosen and immunohistochemical peroxidase-conjugated streptavidin(SP) and reverse transcription PCR method were used to detect the expression of PKGⅡ protein and mRNA.Results: Expression of PKGⅡwas decreased in gastric cancer compared with normal gastric tissues(P<0.05).Result of correlation analysis showed that the expression of PKGⅡ was correlated to the differentiation of gastric cancer cells: the poorer the differentiation,the lower the expression of PKGⅡ.Conclusion: PKGⅡexpression in gastric cancer tissue was decreased and the low expression of PKGⅡmight be associated with the tumorigenesis and the development of gastric cancer.
