摘  要:背景与目的:最近有多项研究表明,Med19基因参与多种肿瘤的形成,如肺癌、乳腺癌和胃癌等。本研究旨在探讨Med19基因表达水平的变化对骨肉瘤Saos2细胞株增殖能力和细胞周期的影响。方法:构建Med19-siRNA及Med19-OE(overexpression)慢病毒载体感染骨肉瘤Saos2细胞,用real time-PCR验证Med19基因的沉默效率及过表达效率,MTT法检测细胞增殖能力,流式细胞仪检测细胞周期,BALB/c裸鼠移植瘤实验检测肿瘤生长情况。结果:转染Med19-siRNA后细胞增殖能力较阴性对照组显著降低(P<0.05),G0/G1期细胞由(36.93±2.96)%上升至(54.95±2.49)%(P<0.05)。转染Med19-OE后细胞增殖能力较阴性对照组显著增加(P<0.05),S期细胞由(13.44±0.76)%上升至(16.50±1.67)%(P<0.05)。BALB/c裸鼠移植瘤实验显示,Med19-siRNA组肿瘤体积及体质量均较对照组显著缩小和降低(P<0.05)。结论:Med19基因沉默可显著抑制Saos2细胞体外的增殖能力,并抑制裸鼠Saos2细胞移植瘤的生长,其抑制增殖作用与诱导G0/G1期阻滞有关。过表达Med19基因可促进Saos2细胞增殖,其促进增殖作用与诱导细胞由G0/G1期向S期过渡有关。Background and purpose:Recently,human Med19 was reported to participate in the carcinogenesis of breast cancer,lung cancer and gastric cancer etc.This study was to investigate the effect of Med19 gene on the proliferation rate and cell cycle of Saos2 osteosarcoma cell.Methods:Med19-siRNA and Med19-OE(overexpression) lentivirus vectors were constructed and then used to transfect the Saos2 osteosarcoma cells.Real time-PCR was employed to assess the gene silencing and overexpression efficacy of these recombinants.The cell proliferation and cell cycle were tested by using MTT and flow cytometry respectively.BALB/c nude mice-transplanted tumors were used to estimate the tumor growth rates.Results:After Med19-siRNA transfection,the cell proliferation rate was significantly lower compared that of the control group(P<0.05).Simultaneously,the proportion of G0/G1 cells increased from(36.93±2.96)% to(54.95±2.49)%(P<0.05).On the contrary,after Med19-OE transfection,the cell proliferation rate was significantly higher than that of the control group(P<0.05),and the proportion of S cells increased from(13.44±0.76)% to(16.50±1.67)%(P<0.05).The tumor volume and weight test showed that the transplanted tumor growth rate of Med19-siRNA group was significantly slower than that of the blank and negative control groups(P<0.05).Conclusion:Silencing of Med19 gene could significantly inhibit the proliferation rate of Saos2 cell and transplanted tumor growth rate.The inhibitory effect may result from the G0/Gl cycle block.While the overexpression of Med19 gene results in the opposite effects.
