摘 要:目的观察Pak1siRNA对胃癌BGC823细胞生长和细胞周期的影响。方法 Westernblot检测Pak1在胃癌细胞中的表达。BGC823细胞转染Pak1siRNA后,MTT分析和细胞计数观察BGC823细胞的生长情况,同时进行细胞周期测定。结果 Pak1在胃癌BGC823细胞中高表达,且能被Pak1siRNA干扰达72.24%。BGC823细胞在转染Pak1siRNA后,生长明显受到抑制(P<0.05),细胞周期在G2/M期发生阻滞,转染Pak1siRNA组G2/M比率[(20.12±1.64)%]较对照组[(8.70±1.69)%,P=0.008]明显升高。结论 Pak1siRNA能够影响细胞周期,从而抑制胃癌BGC823细胞的生长。Objective To observe the effects of Pak1 siRNA to cell growth and cell cycle of gastric cancer cell BGC823.Methods The expression of Pak1 in gastric cancer cell was detected by Western blot.Pak1 siRNA was transfected into BGC823 cell.The growth ability of BGC823 cell was observed by MTT assay and counting cell number,and the cell cycle was detected.Results The expression of Pak1 in BGC823 cell was higher than other gastric cancer cells,and the expression of Pak1 in BGC823 cell interfered by Pak1 siRNA was up to 72.24%.Pak1 siRNA was transfected into BGC823 cell,and then the growth of BGC823 cell was significantly inhibited(P<0.05).The cell cycle exhibited a arrest in G2/M phase,and the G2/M phase percentage of cells by transfecting Pak1 siRNA(20.12%±1.64%)was higher than the control group cells(8.70%±1.69%,P=0.008).Conclusions Pak1 siRNA could affected cell cycle of BGC823 cell,and then inhibited the growth of BGC823 cell.
