摘　　要：目的了解不同人源肝癌细胞系中抑癌基因的甲基化状态,并初步探讨与肝癌发生发展的关系。方法利用甲基化特异性PCR(MSP)技术,选择8个抑癌基因对4种不同类型的肝癌细胞系进行研究。结果通过对4种细胞系的8个抑癌基因甲基化状态分析表明,GSTP1在所有细胞系中均显示甲基化;RASSF1A在SMMC-7721与HepG2中呈现甲基化,而APC在这两个细胞系则是半甲基化状态;P16仅在SMMC-7721中甲基化,SOCS1仅在HepG2中发生甲基化,MGMT仅有HepG2发生半甲基化;RIZ1和DAPK则全部显示未甲基化。结论抑癌基因甲基化谱在不同来源、病理分级和分化程度的肝癌细胞株中各不相同,提示其在基因表达调控中也存在着不同的调节机制,并为体外筛选去甲基化药物诱导肝癌细胞分化模型提供了依据。Objective To investigate the methylation status of tumor suppressor genes in human liver cancer cell lines,also explore the relationship between methylation and the genesis of liver cancer.Methods Four cell lines of human liver cancer were used in this study:SMMC-7721,BEL-7404,HepG2 and TJ3ZX-01.DNA methylation profile of 8 genes (APC,DAPK,GSTP1,MGMT,P16,RASSF1A,RIZ1 and SOCS1) in the 4 cell lines was established by methylation specific PCR.Results GSTP1 methylation was detected in all cell lines.RASSF1A methylation was presented in SMMC-7721 and HepG2,and APC was semi-methylation status in the two cell lines.P16 methylation was only in the SMMC-7721,SOCS1 methylation only in HepG2,and MGMT semi-methylation only in HepG2.RIZ1 and DAPK have no methylation in all 4 cell lines. Conclusions The methylation profile of tumor suppressor genes may be various according to sources,pathological grade and degree of differentiation of HCC cell lines,suggesting that the different regulatory mechanisms are responsible for gene expression.This study provides in vitro cell model to screen demethylation drugs induce differentiation of liver cancer cells.
