甲状腺相关性眼病患者眼眶成纤维细胞高度表达胰岛素样生长因子1受体

摘　　要：目的观察胰岛素样生长因子1受体(IGF-1R)在甲状腺相关性眼病(TAO)患者眼眶成纤维细胞(OFs)的表达及其在TAO发病机制中的作用。方法取确诊为TAO并行眶减压术的患者及外伤眼球摘除术且无其他免疫疾病的正常人眼眶球后结缔组织,培养OFs。通过共聚焦显微镜观察,流式细胞仪检测IGF-1R的表达,并用统计学方法分析比较该受体在TAO患者与正常人之间表达的差异。结果 TAO患者OFs表达IGF-1R为(72.6±10.4)%,正常组为(27.8±10.7)%,两组差异具有统计学意义(P<0.05)。通过共聚焦显微镜观察及定量分析发现,TAO患者OFs的IGF-1R较正常对照组明显表达增多(P<0.05),且IGF-1R主要位于OFs的细胞核周围以及细胞质内。结论与正常人相比,体外培养的TAO患者OFs高度表达IGF-1R。Objective To explore the expression of IGF-1 receptor by orbital fibroblasts(OFs) from patients with thyroid associated ophthalmopathy(TAO) and its effect in the pathogenesis of TAO.Methods Human OFs were harvested and cultured from the connective tissue of patients who were finally diagnosed as TAO undergoing orbital decompression surgery and some ordinary patients without any inflammatory disease undergoing eye enucleate surgery because of trauma.The expression of IGF-1R of OFs was detected by confocal microscopy and flow cytometry,then comparing and analyzing the expression of IGF-1R between patients with TAO and controls by statistical method.Results The expression of IGF-1R by OFs from patients with TAO[(72.6±10.4)%] was significantly different comparing with controls [(27.8±10.7)%](P<0.05).After observation and quantitative analysing by confocal microscopy,we found IFG-1R was over expressed by OFs of TAO patients than normal controls and this receptor was mainly located in the ambient of nucleus and cytoplasm.Conclusions IGF-1R is over expressed by OFs from patients with TAO compared with control donors in vitro.
