SELDI技术在食管癌诊断和治疗监测中的应用

摘  要:目的用蛋白质质谱分析法寻找食管癌诊断、治疗监测以及预后的生物指标。方法采用弱阳离子结合芯片(CM-10)及表面增强激光解吸/电离飞行时间质谱仪(SELDI-TOF-MS)检测食管癌149例、100例正常人的血清蛋白质谱,将获得的图谱采用Ciphergen公司的Biomarker Wizard软件分析。结果食管癌患者与正常人之间有16个差异峰,其中蛋白峰M/Z5622、7750、5322的诊断效能较高。M/Z值为3820的蛋白峰可能为食管淋巴结转移指标,M/Z5622蛋白峰可能与食管癌的远处脏器转移有关。结论该方法可快速、准确检测食管癌,简便易行,是一种有效的筛查手段。且在作为食管癌临床疗效监测及预后方面均有广阔的应用前景。Objective To screen significant biomarkers for early diagnosis,monitoring therapy and prognostic judgement of esophageal carcinoma by using proteomics technique based on mass spectrometry.Methods Surface-enhanced laser desorption/ionization(SELDI) was used to analyze the serum proteins from 149 patients with esophageal carcinoma and 100 healthy persons.All MS spectra generated from SELDI was submitted to Biomarker Wizard software(made by Ciphergen company) for further comparative study.Results 16 differential expressed proteins were identified after comparative analysis between patients with esophageal carcinoma and healthy persons.Among of these,three proteins with the M/Z 5622,7750 and 5322 had high judgement value in diagnosis of esophageal carcinoma.Especially,the protein with M/Z 3820 was revealed to be an important biomarker for lymph node metastasis of esophageal carcinoma and the protein with M/Z 5622 was associated with the distance metastasis of esophageal carcinoma.Conclusions This SELDI technique is very easy to apply in practice and has dramatic benefit in fast and exact detection for early esophageal carcinoma.Therefore,we believe that it would be widely applied for monitoring therapy and prognosis in esophageal carcinoma in future.
