摘 要:目的探讨呼肠孤病毒对乳腺癌细胞的治疗作用。方法比较呼肠孤病毒和表柔比星对MCF-7、BT474细胞的杀灭作用及其干细胞表型CD44+CD24-/low的表达变化(FCM法)。结果表柔比星和呼肠孤病毒均对乳腺癌细胞有杀灭作用。在表柔比星作用下MCF-7和BT474细胞中表达CD44+CD24-/low比例明显升高,为2.5%±0.6%和0.5%±0.1%(P<0.01)。病毒感染后MCF-7和BT474细胞中表达CD44+CD24-/low比例明显下降,均为0(P<0.05)。结论表柔比星对乳腺癌细胞有杀伤作用,但表柔比星可以富集乳腺癌干细胞。呼肠孤病毒对乳腺癌干细胞和癌症细胞同时具有治疗作用,有可能达到关闭癌细胞的目的。Objective To investigate the role of reovirus in breast cancer cells in vitro.Methods Compare the killing results of reovirus and Epirubicin to breast cancer cell lines.Analyze the change of the proportion CD44+CD24-/low expression in MCF-7 and BT474 cells(FCM).Results Both reovirus and Epirubicin could kill breast cancer cells.The proportion of CD44+CD24-/low expression in MCF-7 and BT474 cells treated with Epirubicin improved greatly which were 2.5% ± 0.6% and 0.5%±0.1%(P<0.01).The propotion of CD44+CD24-/low expression in the breast caner cells treated with reovirus decreased to almost none.Conclusions Epirubicin can kill breast cancer cells but it can be used to enrich breast cancer stem cells.Oncolytic reovirus kill breast cancer stem cells and breast cancer cells.Thus reovirus may finally close the breast cancer cells.
