摘　　要：目的探讨shRNA沉默EZH2表达对人肺癌A549细胞增殖和侵袭能力的影响。方法利用RT-PCR方法检测EZH2在人肺癌细胞株中的表达,特异性shRNA沉默EZH2表达后分析NCI-A549细胞增殖和侵袭能力的变化。结果检测14株非小细胞肺癌细胞中EZH2的mRNA表达,80%的细胞株EZH2较正常肺组织高表达;shRNA阻断EZH2表达可显著降低人肺癌A549细胞的增殖和侵袭能力(P<0.05)。结论 EZH2基因沉默能有效抑制人肺癌细胞的增殖和侵袭能力,为深入研究EZH2在非小细胞肺癌中作用提供了研究基础。Objective To investigate the effect of EZH2 knockdown on cell proliferation rate and migration activity in human lung cancer cell line by short hairpin RNA interference.Methods We performed real-time RT-PCR analysis of EZH2 expression in NSCLC cell lines.We also conducted in vitro cell growth assay and matrigel invasion assay by using an EZH2-expressing NSCLC cell line,A549,after knocking down EZH2 expression by an EZH2-specific short hairpin RNA.Results EZH2 mRNA was increased in 80% NSCLC cell lines compared with normal human lung tissue.In the experiment using the A549 cell line,knockdown of EZH2 expression reduced the proliferation rate and invasive activity(P<0.05).Conclusions Short hairpin RNA interference EZH2 can significantly inhibit cell proliferation and invasive activity of A549 cells.It provides a powerful evidence for study of EZH2 role in non-small cell lung cancer.
