中国广东地区汉族人群HLA-B基因多态性与IgA肾病易感性关联研究

摘　　要：目的研究人类白细胞相关抗原(HLA)Ⅰ类B亚型基因多态性与我国广东地区汉族人群IgA肾病易感性的相关性。方法使用基因克隆及聚合酶链反应-测序分型方法(PCR-SBT)对中国广东地区145例IgA肾病患者(IgA肾病组)和137例健康人(对照组)进行HLA-B基因多态性研究,并比较两组的基因型和等位基因的频率,分析HLA-B基因多态性与IgA肾病的相关性。结果 (1)IgA肾病组和健康对照组共检测到103种HLA-B等位基因,其中以HLA-B*400101、HLA-B*4601、HLA-B*1502、HLA-B*1301、HLA-B*5801等位基因频率较高,符合中国广州地区汉族HLA-B等位基因分布特征。(2)HLA-B*5601在IgA肾病组的基因频率明显高于健康对照组(P<0.05),可能与IgA肾病发病相关。结论 HLA-B基因多态性可能与我国南方汉族人群IgA肾病的发病相关,HLA-B*5601可能是IgA肾病的易感基因。Objective To investigate the relationship between polymorphism of HLA-B allele and susceptibility to IgA nephropathy (IgAN) in Hans in Guangdong Region,China.Methods The high resolution gene typing method——PCR-SBT(sequence based typing) and gene colone method were used to investigate 145 IgAN patients and 137 healthy controls in southern Chinese Han population.We studied the association between IgAN and the polymorphism of HLA-B alleles.Results (1) In 145 IgAN and 137 healthy controls,we found 103 HLA-B alleles.Among the alleles,the genetic frequencies of HLA-B * 400101,HLA-B * 4601,HLA-B * 1502,HLA-B * 1301 and HLA-B * 5801 were predominant.The phenomenon was the same with the regular pattern of HLA-B in southern Chinese Han population.(2) In IgAN group,we found that the frequency of HLA-B* 5601 allele was higher(P<0.05),and we considered that HLA-B* 5601 allele may be associated with IgAN.Conclusions It is concluded that HLA-B* 5601 allele is associated with IgAN.
