摘  要:目的:采用 PCR 方法定性检测血中细菌 DNA,快速诊断菌血症的存在。方法:提取全血 DNA(包括细菌 DNA),用 PCR 方法定性检测血中细菌16sRNA 和大肠杆菌β-半乳糖苷酶 DNA。结果:PCR 方法可以检测出血中细菌 DNA,PCR 产物随着细菌 DNA 模板量的增加而增多;采用溶血处理全血可以增加细菌 DNA 检出的灵敏度,全血可冷冻保存;在肝缺血-再灌流大鼠和免疫抑制大鼠血中检出不等量的细菌 DNA;部分脓毒症/发热病人血中可检测到细菌 DNA 的存在。结论:PCR 方法检测全血细菌 DNA 具有简便、快速、灵敏和经济的优点,可用于实验研究及临床菌血症的诊断。To diagnose bacteremia rapidly by PCR method.Methods:After extracting DNA from blood,16sRNA and BG DNA in the blood were analyzed qualitatively by PCR method.Results:Bacteria DNA in the blood could be detected by PCR using 16sRNA and BG primers,the target products of which were increased a- long with the DNA templets.Haemolyzing blood could improve the sensitivity of detecting bacteria DNA in the blood,and the blood could be preserved under frozen condition.Bacteria DNA was detected in the blood of rats with hepatic ischemia or immunity deficiency.Bacterial DNA was also detected in the blood of some sepsis/fever patients.Conclusion:PCR is a simple,rapid,sensitive and inexpensive method to detect bacteria DNA in blood, and it might be used in experimental studies and clinical diagnosis of bacteremia.
