摘  要:目的:观察急性低氧和低氧"习服"时,人肝肿瘤(HepG2)细胞内血管内皮生长因子(VEGF)和低氧诱导因子-1a(HIF-1a)基因表达和蛋白含量的变化。方法:细胞到达低氧"习服"状态后,用 Northern 杂交和Western 杂交检测细胞内 VEGF 和 HIF-1α基因表达以及蛋白含量的变化。结果:急性低氧能够诱导 VEGF、HIF-1α基因表达和胞内蛋白含量升高;而低氧"习服"后再连续低氧48h,细胞内 VEGF 与 HIF-1α的 mRNA 和蛋白含量都恢复到与对照相近的水平。结论:HepG2细胞达到低氧"习服"状态后,耐低氧的能力明显增强,减轻或抑制急性低氧对 VEGF 基因表达的促进作用,其中 HIF-1α可能起着重要的调节作用。Objective;To observe the influence of acute hypoxia and hypoxic'acclimatization'on VEGF and HIF-1a gene expression in HepG2 cells.Methods;HepG2 was cultured in 1% O_2 for 24 hours,then in21% O_2 for another 24 hours,composing a hypoxic training cycle.After 6 cycles,HepG2 cells came to the status of hypoxic 'acclimatization'.During the course of hypoxic training,gene transcription and translation of VEGF and HIF- 1α were estimated with Northern blot and Western blot.Results:Acute hypoxia could induce gene transcription and translation of VEGF and HIF-1α.After hypoxic'acclimatization',gene expression of VEGF and HIF-1a was near normal,while protein content of VEGF HIF-1a fell to control olxlue in the circumstance of 1% O_2 for 48h.Conclusion;When the status of hypoxic'acclimatization'was reached,the hypoxic endurance of HepG2 cells was significantly increased,the hypoxia-induced increment of VEGF gene transcription and translation in HepG2 cells were inhibited in which HIF-1α might play an important role.
