摘 要:目的应用miRNA芯片技术筛选三阴性乳腺癌及癌旁组织差异表达的miRNAs,为进一步阐明其在三阴性乳腺癌发病机制中的作用奠定基础。方法收集8例新鲜的三阴性乳腺癌及其癌旁组织,抽提总RNA进行Hy3荧光标记,miRCURYTMLNA Array(v.16.0)(Exiqon)芯片上进行杂交反应,实时定量PCR验证miRNAs芯片结果的可靠性。结果对芯片进行图像扫描和数据分析,筛选获得17个三阴性乳腺癌相关miRNAs,其中7个miRNAs表达上调,10个miRNAs表达下调;显著上调的有miR-93、miR-328,显著下调的有miR-145、miR-205。结论筛选得到三阴性乳腺癌miRNA差异表达谱,其可能与三阴性乳腺癌的发生发展有关。Objective To screen and identify the miRNA differential expression profile in triple-negative breast cancer by the miRNA array technique.Methods Total RNA was extracted from 8 fresh breast tumor samples and normal adjacent tissues.The miRCURYTM LNA Array(v.16.0)(Exiqon) was used to screen the differentially expressed miRNA.Real-time quantitative PCR was applied to verify the reliability of miRNA array results.Results Compared to normal adjacent tissues,17 miRNAs were differentially expressed in triple-negative breast cancer tissues,including 7 up-regulated and 10 down-regulated miRNAs.The most significantly up-regulated miRNAs were miR-93 and miR-328,while miR-145 and miR-205 were significantly down-regulated.Conclusion We obtained miRNA differential expression profile of triple-negative breast cancer.The profile may be related to the pathogenesis and development of triple-negative breast cancer.
