摘 要:目的观察丹参酮ⅡA对人肝癌SMMC-7721细胞株Ang-2及其受体Tie-2表达的影响,探讨其抗癌的作用机制。方法体外培养肝癌SMMC-7721细胞株,经不同浓度丹参酮ⅡA作用后,MTT方法测定细胞增殖,免疫细胞化学法测定Ang-2及其受体Tie-2表达,ELISA法检测细胞培养液中Ang-2和VEGF水平。结果丹参酮ⅡA对肝癌细胞的生长有明显抑制作用,并呈剂量依赖性。免疫细胞化学检测显示,随着丹参酮ⅡA作用浓度的增大,Ang-2及其受体Tie-2表达下调,其在1.0μg/mL组作用48h的高表达率分别为33.33%、40.00%,灰度值分别为167.43±12.44和169.05±15.59,与空白对照组相比差异有统计学意义(P<0.05);ELISA法检测显示,丹参酮ⅡA作用组细胞培养上清液中Ang-2和VEGF含量明显下降。结论抑制Ang-2及其受体Tie-2表达可能是丹参酮ⅡA抗肝癌作用的机制之一。Objective To investigate the effect of Tanshinone ⅡA on the expressions of angiopoietin-2(Ang-2) and Tie-2 in human hepatocellular carcinoma cell line SMMC-7721. Methods The human hepatocellular carcinoma cell line SMMC-7721 cultured in vitro was treated with Tanshinone ⅡA of different concentrations.Proliferation of the cells was measured by MTT assay,and the expressions of Ang-2 and Tie-2 were detected by immunohistochemical method.ELISA method was used to measure the level of Ang-2 and VEGF in cell culture fluid. Results Tanshinone ⅡA could inhibit the growth of SMMC-7721 cell remarkably in a dose-dependent manner.The immunohistochemical method showed that the expressions of Ang-2 and Tie-2 were down-regulated when the concentration of Tanshinone ⅡA was increased.The high expression ratios of Ang-2 and Tie-2 at 48 h in 1.0 μg/mL group were 33.33% and 40.00%,respectively,and the gray values were 167.43±12.44 and 169.05±15.59,respectively,which were significantly different from the data in normal control group(P<0.05).ELISA method indicated that the contents of Ang-2 and VEGF in cell culture supernatant significantly declined in Tanshinone ⅡA group. Conclusion Inhibiting the expressions of Ang-2 and Tie-2 might be one of the mechanisms of Tanshinone ⅡA in anti-hepatoma effect.
