摘 要:目的 寻找一种更简便、准确的犬下肢深静脉血栓模型构建方法。方法 选择24条雄性比格犬,以犬右下肢股静脉为实验侧,左下肢股静脉为对照侧。二维超声测量静脉管腔直径,彩色多普勒超声定位。麻醉后,超声引导下穿刺股静脉,经动脉鞘送相应粗细的毛刷入犬右下肢股静脉,旋转并拉扯毛刷3.5~4.5cm破坏静脉内膜。术后12h,彩色多普勒超声观察血栓形成情况。结果 超声探头对实验侧股静脉加压,管腔不能被压瘪;彩色多普勒超声示静脉管腔内无回心血流信号;脉冲多普勒超声亦探测不到血流速度,血栓形成。对照侧股静脉血流通畅,探头加压后静脉管腔被压瘪,静脉内无血栓形成。24条犬中22条(91.7%)右下肢股静脉血栓形成,血栓形成长度为(4.0±0.3)cm。结论 超声引导下通过毛刷破坏静脉内膜可成功构建犬下肢深静脉血栓动物模型,操作简便、准确、创伤小、血栓形成率较高。Objective To find a more simple and accurate method for establishing dog lower limbs deep vein thrombosis(DVT) model.Methods Twenty-four male Beagle dogs were included in the study. The right femoral vein was set as the experimental side, and the left femoral vein as the control. The diameter of vein was measured by two-dimensional ultrasound and the location of vein was confirmed by color Doppler ultrasound. After anesthesia, the femoral vein was punctured by the guidance of ultrasound. Brush of appropriate size was placed into the right femoral vein by arterial sheath, then rotated and push-polled for 3.5-4.5 cm to damage the vein intima. After 12 hours, color Doppler flow imaging(CDFI) was used to observe whether there was thrombosis in the vein.Results The standard for thrombosis formation was that the experimental vein can not be flattened by compressing of ultrasound probe, the vein showed no blood flow signals on color Doppler flow imaging and no blood flow velocity on pulse wave Doppler. Femoral vein thrombosis were developed in 22 (91.7%) dogs with an average length of 4.0±0.3cm .Conclusions Lower limbs DVT animal model could be developed successfully by ultrasound-guided damage of the venous intima with brush. This method was feasible, accurate, minimal invasive and with a high successful rate.
