摘  要:目的:探讨蜂斗菜素(petasin)对人神经母细胞瘤细胞SK-N-SH增殖抑制作用的机制。方法:采用罗丹明B(SRB)法检测蜂斗菜素对肿瘤细胞的增殖抑制作用;采用流式细胞术观察蜂斗菜素对SK-N-SH细胞周期和凋亡的影响;采用Hoechst 33258染色法观察蜂斗菜素对SK-N-SH凋亡的影响;蛋白免疫印迹法(WesternBlotting)检测蜂斗菜素对SK-N-SH细胞ERK1/2蛋白磷酸化的影响。结果:蜂斗菜素对SK-N-SH,SGC7901,SPC-A-1,SMMC 7721,Ec9706,SK-N-BE(2)和U251细胞有不同程度的抑制作用。蜂斗菜素使SK-N-SH细胞的周期阻滞于G0/G1期,但不诱导其凋亡,且下调ERK1/2蛋白的磷酸化水平。结论:蜂斗菜素是一种潜在的抗肿瘤天然化合物,其机制为抑制细胞增殖,但不诱导其凋亡。其分子机制可能包括下调ERK1/2蛋白的磷酸化水平。Objective:To study the antiproliferation effects of petasin on human neuroblastoma SK-N-SH cells and the possible mechanisms involved.Methods:The inhibitory effects of petasin on proliferation of tumor cells were detected by SRB assay;the effects of petasin on cell cycle arrest of SKN-SH were measured by flow cytometry;the effects of apoptosis were examined by Hoechst 33258 staining and flow cytometry and the phosphorylation of ERK1/2 of SK-N-SH was determined by western blotting.Results:Compared to that of other cancer cell lines such as SGC7901,SPC-A-1,SMMC 7721,Ec9706,SK-N-BE(2)and U251,the proliferation of SK-N-SH cells was strongly inhibited by petasin.After treated with petasin,the cell cycle of SK-N-SH was arrested in G0/G1 phase,and there was no apoptosis detected either by Hoechst 33258 staining or by flow cytometry analysis.Finally,the phosphorylation of ERK1/2 was decreased by pretreatment with petasin in a dose-dependent way.Conclusion:Petasin is a potential anti-tumor of natural compounds,and shows the strong antiproliferation effect on SK-N-SH cells without inducing apoptosis.The inhibition of ERK1/2 phosphorylation might be involved in the antiproliferation effects of petasin.
