摘 要:采用高盐法、硅珠法、改进的经典酚抽提法三种方法提取人血凝块DNA,通过紫外分光光度仪及琼脂糖凝胶电泳测定DNA的纯度和含量.实验结果表明,硅珠法提取时间快,所提取的DNA含量高;经典酚抽提法操作过程简单,但用有机溶剂提取,污染比较严重;高盐法提取要用大量的血凝块,且提取的DNA含量很不稳定.因此,硅珠法是提取人血凝块DNA的一种比较好的方法.The blood clot s DNA of human beings was extracted respectively by three methods such as higher sodium chloride method,silica gel method and improved phenol-chloroform method,and the degree of purity and the content of the DNA were determined by ultraviolet photometer and agarose gel electrophoresis separately.The results showed that the DNA extraction time of silica gel method was the shortest,and the content of DNA was higher,meanwhile the method of improved phenol-chloroform was the easiest,but the pollution was serious by using organic solvent, whereas higher sodium chlorid method needed a great amount of blood clots and the content of DNA was not stable. Therefore,silica gel method was the bester way to extract the blood clot s DNA of human beings.
