摘 要:目的已有较多研究表明白藜芦醇能抑制多种肿瘤的生长和促进肿瘤细胞的凋亡,观察白藜芦醇对原发性肝癌(简称肝癌)细胞SMMC-7721增殖的影响,探讨其可能的调控机制。方法 MTT法观察不同浓度白藜芦醇(50、100μmol/L)对肝癌细胞SMMC-7721增殖的影响;应用锥虫蓝拒染法检测白藜芦醇对肝癌细胞SMMC-7721的细胞毒性作用;流式细胞术测定肝癌细胞SMMC-7721细胞周期的变化;Western blot法测定细胞周期相关蛋白(Cyclin D1、Cyclin E、P21)变化。结果白藜芦醇呈浓度和时间依赖地抑制肝癌细胞SMMC-7721的增殖(P<0.05)。100μmol/L白藜芦醇干预48 h后,可使G0/G1期肝癌SMMC-7721细胞比例升高,S期细胞比例下降,Cyclin D1和Cyclin E的蛋白表达水平均受到抑制,同时P21蛋白的表达水平上调(P<0.05)。结论白藜芦醇可能通过调控细胞周期来抑制肝癌细胞SMMC-7721的增殖。Objective Resveratrol can inhibit the proliferation of various of tumor cells.This study aims to investigate the effects of resveratrol on the proliferation of hepatoma SMMC-7721 cells.Methods The effects of resveratrol at 50 and 100 μmol/L on the proliferation of hepatoma SMMC-7721 cells were observed by MTT,its toxicity on the cells detected by the trypan blue exclusion method,changes in the cell cycle analyzed by flow cytometry,and the expression levels of the cell cycle related proteins(Cyclin D1,Cyclin E and P21) determined by Western blot.Results The proliferation of SMMC-7721 cells was significantly inhibited by resveratrol in a dose-and time-dependment manner(P<0.05).After 48 hours of 100 μmol/L resveratrol intervention,the transition of the cell cycle from the G0/G1 to the S phase was obviously blocked,the expressions of Cyclin D1 and Cyclin E markedly inhibited,and the expression of P21 significantly up-regulated(P<0.05).Conclusion Resveratrol may inhibit the proliferation of SMMC-7721 cells by regulating the their cell cycle progression.
