摘  要:目的探讨内毒素对老龄大鼠脑细胞凋亡的影响。方法观察正常以及门静脉注射内毒素后24月龄、6月龄大鼠血浆内毒素与TNF-α水平的变化;体外观察内毒素和TNF-α对脑细胞胞浆游离钙的影响,通过流式细胞仪检测脑细胞凋亡情况。结果正常与注射内毒素后24月龄大鼠血浆内毒素水平均显著高于6月龄大鼠(t=3.684,P<0.05;t=4.583,P<0.05),而TNF-α明显升高仅见于注射内毒素后(t=7.766,P<0.001);TNF-α可使脑细胞胞浆游离钙明显升高(t=2.668,t=73.589,t=4.641,P<0.05);发现门静脉注射内毒素的24月龄、6月龄大鼠均发生脑细胞凋亡(50.53%,29.60%)。结论老龄大鼠血浆内毒素升高,致使脑细胞胞浆游离钙升高、细胞凋亡发生,这可能与细胞因子改变脑细胞微生态环境有关。Objective To study the effect of endotoxin on brain cell apoptotic in aging rats.Methods Endotoxin and TNF-α content of plasma were determined in normal and after endotoxin infection of 24 months old and 6 month old rats.Effect of endotoxin and TNF-α on free Ca2+ in brain cells were observed in vitro,and apoptosis of brain cells were examined by FCM.Results The level of endotoxin in plasma were much higher in 24 months old rats than those of 6 months old rats in both normal and after endotoxin injection groups,but TNF-α was increased significantly only after endotoxin injection.TNF-α was elevated significantly free Ca2+in brain cells,and after endotoxin injection via portal vein apoptosis of brain cells were developed.Conclusions Increased plasma endotoxin toxin and TNF-α in 24 months old rats contributed to elevation of Ca2+ in brain cells and apoptosis,and the mechanism might involves changes of brain microecological environment induced by altered cytokine.
