摘 要:目的分析非小细胞肺癌中MicroRNA let-7与基质金属蛋白酶2(MMP-2)的表达及临床意义,探讨其在肺癌发生及发展中的作用及相关性。方法采用免疫组化和Western blot检测46例肺癌及配对的癌旁组织中MMP-2的表达,用实时定量PCR检测46例肺癌组织及配对的癌旁组织中let-7和MMP-2的表达,Spearman等级相关分析检测两者的相关性。结果免疫组化结果示46例非小细胞肺癌组织中MMP-2阳性表达30例(65.2%)。MMP-2表达与患者性别、年龄、肿瘤类型无关(P>0.05),与肿瘤临床分期有关(P<0.05),与淋巴结转移有关(P<0.05)。Western blot示MMP-2在肺癌中表达高于癌旁正常组织。实时定量PCR示let-7在非小细胞肺癌组织中与配对的癌旁组织比较表达降低的为29例(63.0%),MMP-2高表达的为31例(67.4%)。46例肺癌组织中,let-7与MMP-2表达负相关(rs=-0.433,P<0.05)。结论 MMP-2及let-7在肺癌中的表达存在异常,let-7作为抑癌因子它的低表达可能与MMP-2的高表达有关,两者之间可能存在某种协调关系。Objective To analyze expression of MicroRNA let-7 and matrix metalloproteinases 2(MMP-2)and their clinical significance and to discusses their role in non-small cell lung cancer(NSCLC)and correlation during lung carcinogenesis.Methods MMP-2 expression was determined using immunohistochemical SP and Western blot,and clinicopathological features were analyzed.Realtime quantitative PCR analysis was conducted to detect let-7 and MMP-2 expression,and the relationship between let-7 and MMP-2 was analyzed.Results The immunohistochemisty results showed among 46 NSCLC,MMP-2 was stained positively in 30 cases(65.2%).Expression of MMP-2 in NSCLC did not show significant difference between pathological type,age and sex(P>0.05).The positive expression of MMP-2 was associated with tumor stage and lymph node metastasis(P<0.05).Western blot analysis showed that MMP-2 protein expression was significantly higher in lung cancer tissue,compared with those in paraneoplastic tissue.Realtime quantitative PCR analysis showed that let-7 lower expression for 29 cases(63%);MMP-2 higher expression for 31 cases(67.4%).Spearman′s correlation revealed that MMP-2 expression was correlated negatively with let-7 in 46 lung cancer specimens(rs=-0.433,P<0.05).Conclusions Our result indicated that let-7 and MMP-2 maybe involved in lung cancer and may imply coordination in lung tumorigenesis.
