摘 要:目的探讨FIZZ1/RELMα对大鼠动脉内皮细胞(SVARECs)的血管新生能力的影响及其机制。方法用贴壁法培养大鼠主动脉内皮细胞,采用Matrigel小管形成实验来评价不同浓度(10-8、2×10-8mol/L)的FIZZ1对大鼠主动脉内皮细胞血管新生的作用;采用全基因表达谱检测FIZZ1对主动脉内皮细胞信号通路活性的影响,筛选出显著性差异表达的基因。结果用1×10-8mol/L、2×10-8mol/L的FIZZ1刺激大鼠主动脉内皮细胞后,发现处理组SVARECs的体外血管新生数目均明显高于正常对照组,且呈剂量依赖[分别为(26.5±4.33)个/HP和(34.7±5.24)个/HP vs.(19.2±3.65)个/HP,P均<0.01]。用FIZZ1刺激大鼠主动脉内皮细胞后,全基因表达谱检测提示有937条显著性差异表达基因,其中上调基因440条;下调基因497条。结论 FIZZ1可促进大鼠主动脉内皮细胞血管新生,其机制信号通路Gng8、Atg9a、Gdf6等基因显著性表达有着密不可分的关系。Objective To study the angiogenic ability of FIZZ1 on cultured aortic rat endothelial cells(SVARECs)and the potential mechanism.Methods SVARECs were cultured in RPMI-1640 supplemented with 10% FBS.Matrigel assay were used to detect the angiogenic ability of the SVARECs after stimulation by FIZZ1(1×10-8,2×10-8 mol/L).The agilent rat microarray containing 41 000 genes/ESTs were used to screen the differentially expressed genes of the SVARECs after stimulation by FIZZ1.Results In a dose-dependent manner(1×10-8,2×10-8 mol/L)FIZZ1 markedly increased the angiogenic ability of SVARECs[(26.5±4.33)/HP and(34.7±5.24)/HP vs.(19.2±3.65)/HP,all P<0.01].Towards the screening and analysis of the important differently expressed genes in the SVARECs after stimulation by FIZZ1,there were 440 genes(Gng8,Atg9a,Gdf6,etc.)with upregulated expression and 497 genes(Hbb-b1,Camk1g,etc.)with downregulated expression in the experimental group.Conclusions FIZZ1 could stimulate SVARECs in angiogenesis through activation of regulation of actin cytoskeleton pathway.
