摘  要:目的:观察血红素氧合酶-1(heme oxygenase-1,HO-1)抑制剂对高氧暴露早产鼠视网膜血管内皮生长因子(vascular endothelial growthfactor,VEGF)和色素上皮衍生因子(pigment epithelium-derived factor,PEDF)表达的影响。方法:将144只3日龄早产大鼠随机分为A高氧组、B干预组、C空气组、D空气+舒血宁组,以免疫组织化学法检测各组早产鼠的视网膜HO-1、PEDF和VEGF表达水平。结果:C、D组HO-1、PEDF、VEGF均呈低表达,未见或罕见突破内界膜的内皮细胞核;与C、D组相比,A组突破内界膜的内皮细胞核明显增多,HO-1、PEDF及VEGF表达的表达均显著增加,(P分别<0.001,0.001,0.001),与A组相比,B组突破内界膜的内皮细胞核明显减少,HO-1、PEDF及VEGF的表达均显著下降(P分别<0.05,0.001,0.05);视网膜PEDF的表达与VEGF的表达、视网膜PEDF的表达与突破视网膜内界膜内皮细胞核数呈负相关、VEGF、HO-1的表达与突破视网膜内界膜内皮细胞核数呈正相关(P分别<0.01,0.01,0.05)。结论:高氧暴露早产鼠视网膜内可观察到HO-1、VEGF和PEDF的高表达,舒血宁通过下调高氧暴露早产鼠视网膜HO-1、VEGF和PEDF的表达,对高氧性视网膜病有一定的保护作用。Objective:To observe the effect of heme oxygenase-1(HO-1)inhibitor on the expression of vascular endothelial growth factor(VEGF)and pigment epithelium-derived factor(PEDF)in the retina tissue of preterm rats exposed to hyperoxia,and explore the role of HO-1 inhibitor in hyperexia-induced retina injury of preterm rats.Methods:one hundred and forty-four three-day-old preterm rats of standard SD were randomly assigned to hyperoxia group,hyperoxia+gingkgo injection group,room air group and room air+gingkgo injection group.The expression of HO-1,VEGF and PEDF in the retinas were measured by immunohistochemical techniques.Results:The expression of HO-1,VEGF and PEDF were low in the room air group and room air+gingkgo injection group,and the nubers of the endothelial nucleis breaked the membrana limitans interna were rare in these two groups.The nuber of the endothelial nucleis breaked the membrana limitans interna and the expression of HO-1,VEGF and PEDF in the hyperoxia group were obviously increased compared with up two groups (respectively P<0.001,0.001,0.001).Meanwhile,The nuber of the endothelial nucleis breaked the membrana limitans interna and the expression of HO-1,VEGF and PEDF in hyperoxia+gingkgo injection group were significantly decrease compared with hyperoxia group (respectively P<0.05,0.001,0.05).The nuber of the endothelial nucleis breaked the membrana limitans interna and the expression of HO-1,VEGF and PEDF were positive correlatio(nrespectively P<0.01,0.01,0.05).Conclusion:This study shows that expression of HO-1,VEGF and PEDF was significantly increased when exposed to hyperoxia,gingkgo injection could down-regulate the expression of HO-1 VEGF and PEDF gene in the retinas of hyperoxia-induced preterm rats.
