摘 要:目的探讨阿托伐他汀对阿尔茨海默病(AD)模型小鼠学习、记忆功能及Klotho蛋白的影响。方法采用Aβ1-40脑内注射的方法构建AD小鼠模型,将小鼠分为4组:模型组、生理盐水组、阿托伐他汀组及加兰他敏组;用Morris水迷宫实验检测学习及记忆功能,用酶联免疫法检测小鼠海马组织Klotho蛋白及胰岛素的含量。结果与模型组比较,阿托伐他汀组逃避潜伏期明显缩短,跨越平台次数显著增加(P<0.05);Klotho蛋白及胰岛素水平的表达显著增加(P<0.05)。结论阿托伐他汀可改善AD小鼠的学习及记忆功能,其机制可能与其对Klotho基因表达的上调有关。Objective To investigate the effects of atrovastain on the study,memory function and the expression of Klotho protein in mice with Alzheimer's disease(AD).Methods AD model was established by Aβ 1-40 injection.All mice were divided into four groups: model group,physiological saline group,atrovastain group and galanthamine group.Morris trial was used to assess the study and memory function,and ELISA was used to detect the expression level of Klotho protein and insulin.Results Atrovastain can shorten the escape latency,and enhance step platform times(P < 0.05).In addition,atrovastain can improve the expression of Klotho and the level of insulin.Conclusions Atrovastain may improve the study and memory function in mice with AD through upregulating Klotho gene.
