钙蛋白酶10 SNP43(G/A)基因多态性与甘肃东乡族人群2型糖尿病的关系

摘　　要：目的研究中国甘肃东乡族钙蛋白酶10基因单核苷酸多态性(SNP43)(G/A)位点多态性,探讨其与胰岛素抵抗、2型糖尿病发病的关系。方法应用聚合酶链式反应-限制性片段长度多态性技术对随机选取的东乡族2型糖尿病患者38例和同期体检的85名东乡族对照组钙蛋白酶10基因SNP43(G/A)位点进行多态性分析。采用氧化酶法或放免法测定血糖、胰岛素及血脂水平。结果 2型糖尿病组和对照组基因型及等位基因的频率比较差异无统计学意义;在2型糖尿病组钙蛋白酶10基因SNP43的G/G型与G/A型及A/A型携带者空腹胰岛素、胰岛素抵抗指数相比较,差异均有统计学意义(P<0.05),而空腹血糖、甘油三酯、总胆固醇、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇在2型糖尿病组和对照组3种基因型携带者间相比较,差异均无统计学意义。结论钙蛋白酶10 SNP43(G/A)在中国甘肃东乡族人群中存在基因多态性,该基因多态性与甘肃地区东乡族人群胰岛素抵抗相关。Objective To investigate the distribution of calpain 10 SNP43（G/A） polymorphism in people of Dongxiang nationality in Gansu Province and its association with insulin resistance, type 2 diabetes mellitus（T₂DM）.Methods Polymerase chain reaction-restriction fragment length polymorphism method was used to test the polymorphism of calpain 10 SNP43（G/A）.A total of 38 individuals from T₂DM and 85 normal control subjects were admitted.Plasma glucose, insulin,lipid levels were measured by biochemical technique.Results There had no significant difference of genotype and allele gene frequency between T₂DM group and control group.In T₂DM group,there were significant differences in fasting insulin,Insulin Resistance Index,for calpain 10 SNP43 G/G genotype subjects,calpain 10 SNP43 G/A and A/A genotype subjects （P &lt;0.05）.There were no significant differences in fasting plasma glucose,triglyceride,highdensity lipoprotein cholesterol,low-density lipoprotein cholesterol,total cholesterol for T₂DM group and control group among the three genotypes subjects.Conclusion Calpain 10 SNP43 （G/A） polymorphism exists in the Dongxiang nationality population in Gansu Province and it is correlated to their insulin resistance.
