摘 要:目的研究opiorphin在氟代二甲基亚砜(DMSO-d6)溶液中的构象。方法以固相多肽合成方法合成opiorphin,用2D NMR技术研究opiorphin在DMSO-d6溶液中的构象,记录COSY、TOCSY和NOESY共振谱用于构象分析。结果利用2D TOCSY确定了opiorphin多肽氨基酸残基的自旋系统,通过解析NOESY谱中的NOE相关点,完成了对其序列中主链和侧链质子的全部归属,并分析得到肽链的~1H的化学位移和质子间的NOE。三维结构计算表明opiorphin在DMSO-d6中为规划则卷曲形式存在。结论柔性无规则卷曲的柔性特征可能是opiorphin与其受体结合前的最佳状态。Objective To investigate the conformation of opiorphin in deuterated dimethyl sulfoxide(DMSO-d<sub>6</sub>) solution.Methods Opiorphin was synthesized by solid phase peptide synthesis.Its solution in DMSO-d<sub>6</sub> was examined by 2D NMR technique.The COSY,TOCSY and NOESY spectra were recorded for the analysis of the conformation.Results The sequence-specific assignment of spin systems was obtained by 2D TOCSY spectra,and the complete assignment of proton resonances for backbone and side chain were achieved by NOEs correlation in NOESY.The chemical shifts of <sup>1</sup>H and NOE suggested that opiorphin existed mainly as random coil in DMSO-d<sub>6</sub>.Conclusion This random coil conformation may be suitable for binding its receptor.
