MICA在胃癌中的表达及其临床意义的研究

摘　　要：目的研究MICA分子在胃癌组织中的表达与胃癌生物学行为的关系。方法取2010年9月至2011年1月青岛大学医学院附属医院普外科行胃癌根治术患者的胃癌组织及胃正常组织标本40例,一式两份,一份用作病理分析,另一份用作RT-PCR分析,患者在手术前均未行放化疗,临床病理资料完整,肿瘤分期按照2010年NCCN颁布的方案实施。所有病例均经术后病理学检查证实为胃癌;用荧光定量RT、-PCR法分别测定胃癌组织和胃正常组织中MICA的表达情况;汇总胃癌组织的临床病理资料,说明胃癌组织MICA的表达与胃癌生物学行为的关系。结果①荧光定量PCR结果显示胃癌组织中MICA的表达(1.624±:1.380)明显高于胃正常黏膜组(1.072±0.524)(p<0.05);②MICA高表达的胃癌组织的生物学行为较良,MICA低表达的胃癌组织的生物学行为较恶。结论①胃癌组织中高表达MICA分子;②MICA的表达水平与胃癌组织的生物学行为有关系。Objective To investigate the relation between the expression of MICA in the tissue of gastric cancer and the biological behaviour of gastric cancer.Methods The tissue samples of gastric cancer tissue and gastric normal tissue in 40 patients with gastric cancer who underwent radical gastrectomy at general surgery in the Affiliated Hospital of Medical College Qingdao University from September 2010 to January 2011 were selected.AU the samples were collected within 30 min after operation,which were divided into 2 portions with one portion for pathology analysis,another for succedent RT-PCR analysis and all patients did not undergo radiotherapy and chemotherapy.Clinical and pathological information in all patients were all integrated.The tumor stage was practiced according to NCCN in 2010.All cases were confirmed as gastric cancer by pathology after operation.The expression of MICA in gastric cancer tissue and normal gastric tissue were determined by RT-PCR analysis.The clinical and pathological information of gastric cancer tissue was collected to explain the relation between the expression of MICA in gastric cancer tissue and biological behaviour of gastric cancer.Results ①The results of fluorescent quantitative PCR demonstrated that the expression of MICA in gastric cancer tissue(1.624±1.380) were obviously higher than those of gastric normal tissue(1.072±0.524)(P<0.05);②The biological behaviour of gastric cancer tissue that highly express MICA is better and that lowly express MICA is more malignant.Conclusions The MICA have a high expression in gastric cancer tissue and the expression level of MICA is related to biological behaviour of gastric cancer tissue.
