摘 要:以纤维素粉为唯一碳源,从棉籽壳中筛选出能分解纤维素(CMC)的霉菌,利用常规方法鉴定为绿色木霉,通过紫外线诱变提高分解纤维素的能力,采用水解圈和滤纸崩溃法进行初筛,摇瓶培养复筛的方法,筛选出一突变绿色木霉(T118),降解纤维素的能力提高了1倍,CMC酶活性提高了50%,C1酶活性提高了40%,还对该突变株产酶条件进行了研究,得出最适条件为:稻草粉:麸皮=4:1,发酵时间为5d,发酵温度为30℃,该酶最适反应PH为6,最适反应温度为50℃。此突变株易培养,有利于工业生产,具有重大的实用价值。The mould of cellulose-decomposing microorganis is isolated from with cellulose-flour as the solecarbon source.The mould was identified as Trichoderma Viride by the routine method,the ability of productingcellulase increased after Trichoderma Viride was mutated with ultraviolet light,A mutant strain TfrichodermaViride (T118)was obtained by initial screening with the method of distinct zones and filter paper collapses,cellulase activity measurement of culture filtrate,The ability of cellulose-decomposing was raised one time.Theactivities of CMCase and C1ase increased 50%and40%,the optimum conditions for producing cellulase wereproportion of straw to wheat 4:1,time duration of fermentation5d,temperature30℃,The optimum pH and tem-perature for enzymatic catalysis were 6 and50℃ respectively.This mutant strain nurture easily and bendfiesindustrial production,the strain has a applied value.
