摘 要:目的筛选合适的大豆磷脂以及氢化磷脂配比混合物作为膜材,利用主动载药法制备氧化苦参碱脂质体。方法将商品大豆磷脂和氢化磷脂以一定的配比制备膜材,采用薄膜分散-主动载药法进行制备。并考察在室温挤出时的难易程度和挤出后脂质体的形态及包封率。结果主动载药法制备的氧化苦参碱脂质体包封率较高,在选择一定的磷脂配比后,主动载药的包封率质量分数可在80%左右,且挤出后的脂质体粒径较圆整。结论筛选合适的膜材在常温下可进行挤出整粒操作,采用主动载药法制得氧化苦参碱脂质体包封率较高。Objective To screen suitable hybrid membrane material composed of soybean phospholipid and hydrogenated soybean phospholipid to prepare liposomal oxymatrine by pH-gradient drug loading method. Method Different ratio of soybean phospholipid and hydrogenated soybean phospholipid was composed for preparing liposome by membrane-dispersing and pH-gradient drug loading method. The difficulty of extrusion of hybrid membrane liposome at room temperature was investigated. The morphous of the liposome after extruding was observed and the encapsulation efficiency was determined. Results The encapsulation efficiency is high by this method. A suitable composition of phospholipid resulted an encapsulation efficiency of 80%.The liposome particle was round and had a good shape. Conclusions The method employed is suitable for preparing liposomal oxymatrine on industrial scale.
