摘  要:为了研究高压氧 (hyperbaric oxygen,HBO)暴露对体外培养的大鼠神经元生长的影响以及血小板源性生长因子 -BB(platelet derived growth factor-BB,PDGF-BB)和阿魏酸钠的保护作用 ,本研究采用原代培养大鼠大脑皮层和海马神经元 ,在5 0 0 k Pa高压氧中暴露 3 0 m in和 60 min,MTT法检测神经元的存活率 ,并观察神经元的形态变化。此外 ,用形态学观察结合图像分析研究 PDGF-BB的促神经元生长作用。结果显示高压氧暴露后 ,神经元存活率明显下降 ,且随暴露时间延长 ,存活率下降越显著 ,神经元形态也出现明显的损伤性改变。预先给予 PDGF-BB可促进神经元胞体和突起的生长 ,使存活率明显提高 ,阿魏酸钠则没有显著影响。上述结果表明过度高压氧暴露能损伤神经细胞 ,降低神经元的存活率 ,PDGF-BB可促进神经元的生长 ,起神经保护作用 ,从而提高存活率。To investigate the influences of hyperbaric oxygen (HBO) on survival of cultured neurons and the protection of platelet-derived growth factor (PDGF-BB) and sodium ferulate (SF), hippocampal and cortical neurons of neonate rat were cultured for 10 days, then were exposed to 500 kPa oxygen. The survival rates of neurons were measured with MTT, and the changes of form were also evaluated. In addition, the growth-promoting abilities of PDGF-BB on neurons were investigated. The results showed that the survival rate of neurons decreased significantly after being exposed to 500 kPa HBO for 30 and 60 min, and the neurons presented notablely damaged appearances. PDGF-BB could promote the growth of body and neurites of neurons, and survival of neurons, but SF had no effects. The results indicate that excessive HBO exposure can damage the neurons, and decrease their survival. PDGF-BB had neurotrophic effects on neurons, and could promote the growth of neurons, therefore the survival of neurons exposed to HBO was improved. However SF had not effects.
