摘  要:二脂酰甘油酰基转移酶1(acylCoA:diacylglycerol acyltransferase,DGAT1)基因是影响奶牛产奶性状的主效基因之一,其第八外显子上的第232位的赖氨酸转化为丙氨酸(K232A)的错义突变是影响产奶量、乳脂和乳蛋白含量的因果突变。本研究利用GenBank中已公布的牛DGAT1基因DNA序列,于K232A位点两端设计引物,采用PCR-RFLP方法鉴定来自新疆和江西的3个中国荷斯坦母牛群体共454个个体DGAT1基因K232A突变位点的基因型,使用SAS9.0软件分析供试群体基因型分布及其与产奶量的相关性。研究结果表明:K等位基因为441bp的PCR扩增片段,A等位基因为完全切开的202bp和239bp两片段。新疆1、新疆2和南昌金牛3个荷斯坦母牛群体在该位点上K等位基因的频率分别为51.9%、42.4%和27.2%;A等位基因的频率分别为48.1%、57.6%和72.8%。DGAT1基因K232A突变位点与三个母牛群体日平均产奶量的相关性分析发现在3个群体中KK型个体产奶量均低于AA型个体,但不同基因型个体间产奶量差异在统计学上没有显著差异。本研究为进一步探讨DGAT1基因在中国奶牛群体中的遗传效应及分子标记辅助选择培育良种奶牛提供参考。acylCoA,diacylglycerol acyltransferase(DGAT1),is one of the major effect genes for traits of cow milk production.The missense mutation which caused by the substitution of a lysine residue with a alanine residue at 232(K232A) on its exon 8 is the causative mutation for milk yield,milk fat and protein contents.In this study,according to bovine DGAT1 genomic DNA sequence published in GenBank database,we designed the primers for amplifying the fragment of DGAT1 gene at both loci of K232A and determined the genotype of each individual at K232A locus in 454 Holstein cows collected respectively from three farms in Xinjiang and Jiangxi province by PCR-RFLP.Then distribution of each allele and its association with milk yield in these three Holstein cow populations were analyzed bySAS9.0 software.There sults showed that allele K was the 441 bp PCR amplification product,allele Acontained two fragments of 202bp and 239bp which were from PCR products completely digested.the frequency at this locus of all ele Kin three Holstein cows populations of Xinjiang1,Xinjiang 2 and Nanchang Jinniu was 27.2%,51.9% and 42.4%,respectively,and that of the allele A was 72.8%,48.1% and 57.6%,respectively.Association analysis of the K232A mutation locus of DGAT1 gene with daily average milk yield of this three populations demonsrtated that the milk yield of individuals of KK type had lower than that of AA one in these 3 populations,but the milk yield of individuals in different genotype was not statistically significant difference.The results of this study would provide the reference for further investigating the genetic effect of DGAT1 in Chinese Holstein cow populations and cultivating the well-bred cow breeding through molecular marker-assisted selection.
