摘 要:构建了4种扇贝扩增片段长度多态性(AFLP)的分析体系,对DNA提取、双酶切反应、连接反应、预扩增反应、选择性扩增反应和银染等条件进行了优化。结果表明:用EcoRⅠ/MseⅠ双酶切,核心序列加3个选择性碱基的选择性扩增引物,E32-M49和E32-M62两个引物组合,以及20μL选择性扩增体积,可得到十分稳定的结果。所得产物经电泳和银染后可获得条带清晰、背景干扰小的图像。该体系的构建为AFLP技术在扇贝相关研究中的应用提供了依据。DNA extraction,double enzymes digestion and adapter ligation reaction,preamplification and selective amplification reactions and argent dying were optimized to establish AFLP(Amplified Fragment Length Polymorphism) analysis system for four scallop species.It was proved that quite steady results were achieved by using EcoRⅠ/MseⅠ double enzymes digestion,three selective base primers,two of informative primers and 20 μL volume of selective amplification.The pictures of the production by electrophoresis and argent dying were very well,with the high resolution bands and less disturbed.The establishment of the system provided a foundation for the application of AFLP techniques to the relative interspecies research of scallop.
