新疆Kaposi肉瘤患者感染的人类疱疹病毒8型ORF75基因多态性研究

摘　　要：目的通过对人类疱疹病毒8型(HHV-8)ORF75基因多态性研究,初步探讨其与新疆Kaposi肉瘤的相关性。方法对25例新疆Kaposi肉瘤患者石蜡包埋组织进行HHV-8 DNA抽提、扩增及双向测序,使用CLUSTALW软件和PHYLIP软件包对ORF75基因进行核苷酸多态性分析。结果①25例新疆Kaposi肉瘤患者有21例HHV-8感染阳性,阳性率为84%,其中7例艾滋病相关型Kaposi肉瘤(AIDS-KS)患者HHV-8感染均为阳性。②新疆Kaposi肉瘤患者HHV-8 ORF75基因核苷酸具有多态性,存在462、618、647三个主要突变位点。结论人类疱疹病毒8型ORF75基因多态性与新疆Kaposi肉瘤具有相关性。Objective To discuss the correlations between Kaposi's sarcoma in Xinjiang and HHV-8 ORF75 gene polymorphism. Methods HHV-8 DNA was amplified by nested PCRand sequenced which had been extracted from paraffin-embedded tissue example of 25 patients with Kaposi's sarcoma in Xinjiang.Then the polymorphism analysis was carried out by using the CLUSTALW program and the PHYLIP package.Results Of the 25 patients with Kaposi's sarcoma,21 patients were HHV-8 positive(84%) and the 7 cases with AIDS-KS were all turned out to be HHV-8 positive.HHV-8 ORF75 gene locus nucleotide polymorphisms has been identified,and three main mutations detected in 462,618 and 647.Conclusion HHV-8 ORF75 gene polymorphism has a close relation with Kaposi's sarcoma in Xinjiang.
