摘 要:目的:探讨凋亡抑制基因survivin和cox-2、bcl-2在非小细胞肺癌中的表达及与临床病理学特征的关系。方法:采用免疫组织化学SP法检测30例非小细胞肺癌及相应的癌旁组织中survivin、cox-2和bcl-2蛋白的表达。结果:(1)survivin、bcl-2蛋白在癌旁组织中未见阳性表达,在肺癌组织中的阳性表达率为76.7%、60%,其阳性表达率与肿瘤大小、组织类型、组织学分级、淋巴结转移和TNM分期均无关(P>0.05)。肺癌和相应的癌旁组织中cox-2蛋白的阳性率分别为66.7%、10%。肺癌组明显高于癌旁组(P<0.001)。其阳性表达率与肿瘤大小、组织类型无关(P>0.05);而与组织学分级、淋巴结转移、TNM分期有关。(2)在非小细胞肺癌中Survivin与cox-2的表达呈正相关。survivin与bcl-2表达无关。结论:survivin、cox-2和bcl-2基因的过表达可能对非小细胞肺癌的发生和发展起重要作用,它们可能为肺癌的早期诊断和基因治疗提供新的靶点。Objective:To investigate the expression of the inhibitor apoptosis gene survivin and cyclooxygenase-2(cox-2),bcl-2 in lung cancer tissue and their correlation with clinicopathological features.Methods:The expression of survivin,cox-2 and bcl-2 protein was detected by immunohistochemical(SP technique) in cancer tissue and tumor-adjacent tissue in 30 cases of lung cancer.Results:(1) The positive rates of survivin and bcl-2 protein in cancer tissue were 76.7%(23/30)、60%(18/30),and no expression in tumor-adjacent tissue.The positive rate in lung cancer was not related with size of tumor and histological type clinic pathological parameters including histological grade,lymphnode metastasis and TNM staging(P>0.05).The positive rates of cox-2 in cancer tissue and tumor-adjacent tissue were 66.7%(20/30)、10%(3/30).There was highly significant difference between the cancer tissue and tumor-adjacent tissue(P< 0.001).The positive rate in lung cancer was correlated with clinic pathological parameters including histological grade,lymphnode metastasis and TNM staging(P<0.05)but no correlated with size of tumor and histological type(P>0.05).(2) The expression of survivin was positively correlated with that of cox-2.There was no correlation between the positive rates of survivin and that of bcl-2.Conclusion:The specific up-regulation of survivin gene and cox-2 、bcl-2 gene in lung cancer tissues suggested that they may play an important role in the process of carcinogenesis and may be indentified as a defining diagnostic marker and a potential therapeutic target in lung cancer.
