摘  要:目的:探讨人参茎叶皂甙(GSL)对人肝癌细胞增殖的抑制作用及其机制。方法:人肝癌细胞系 SMMC 7721在体外培养条件下,经浓度为200 gg/ml 的 GSL 作用3天后,进行如下检测:应用流式细胞仪检测对照组和 GSL 组人肝癌细胞细胞周期频数分布及细胞周期蛋白 D_1(Cyclin D_1)表达水平;运用原位杂交技术检测其 N-ras 基因 mRNA 转录水平,通过图像分析仪测量其胞浆内阳性杂交颗粒的平均吸光度值进行半定量分析。结果:GSL 组 S 期细胞数明显小于对照组(P<0.01);Cyclin D_1蛋白表达及 N-ras 基因 mRNA转录亦明显较对照组降低(P<0.0)。结论:人参茎叶皂甙通过抑制 N-ras 基因 mRNA 转录,进而 Cyclin D_1蛋白表达下调,从而使较多的人肝癌细胞被阻止于 G_1/S 检查点(checkpoint)。To study the inhibitory effect of ginsenosides from stems and leaves(GSL)on proliferation of human hepatocarcinoma cell line SMMC 7721 and its mechanism.Methods:Human hepatocarcinoma cells were treated in vitro with 200 μg/ml GSL for 3 days, then frequency distribution of cell cycle and expression of Cyclin D_1 protein were tested by flow cytometry.Transcription of N-ras gene was measured by in situ hybridization assay and the mean absorbance of positive granules in cytoplasm was analyzed semi-quantitatively by image analysis.Results:The S phase cell count in GSL group was significantly lower than that in the control group(P<0.01),and the expression of Cyclin D_1 protein and transcription of N-ras gene were also significantly down-regulated(P<0.01).Conclusion:GSL could inhibit transcription of N-ras gene,and consequently down-regulate expression of Cyclin D_1 protein to let more hepatocarcinoma cells staying at G_1/S checkpoint,so as to inhibit the proliferation of human hepatocarcinoma cell line.
