摘  要:目的?观察γ氨基丁酸对可溶性淀粉样前体蛋白(sAPP)释放的影响。方法:雄性SD大鼠断头后取海马于0~4℃ Krebs-Ringer液中作快速振动切片,切片在37℃预孵育后,置于含或不含γ氨基丁酸Krebs-Ringer液中孵育后于15000×g 离心15min取上清,应用二辛可宁酸法测定上清总蛋白含量;释放入孵育液中的sAPP以Western印迹法测定。结果:正常 孵育的海马脑片能够分泌基础量的sAPP,γ氨基丁酸(1.0×10-9~1.0×103μmol/L)可导致海马脑片释放sAPP减少,并且 没有导致新蛋白的产生。结论:γ氨基丁酸在一定浓度范围内能够抑制海马脑片分泌sAPP。Objective:To study the effects of γ-GABA on the release of the soluble amyloid precursor protein(sAPP).Methods:Male Sprague-Dawley rats were decapitated and hippocampus was isolated at4℃,then hippocampal slices were quickly prepared in the medium of Krebs-Ringer buffer at0-4℃by a vibration chopper.After the slices were preincubated in Krebs-Ringer buffer at37℃,they were stimulated with various concentrations of γ-GABA(1.0×10 -9 -1.0×10 3 μmol/L)in Krebs-Ringer buffer.The supernatant was removed from the solution after centrifugated at15000×g.Quantities of total pro-tein were measured by bicinchoninic acid(BCA)assay.The sAPP released into the incubation medium were assayed by West-ern blot.Results:γ-GABA increased sAPP release in the incubated hippocampal brain slices.However,it failed to produce special protein.Conclusion:γ-GABA within certain concentration range has inhibitory effects on the sAPP release.[
