摘  要:目的为探讨CyclinD1、CDK4基因及蛋白在食管癌发生中的意义及其相互间的作用。方法采用LSAB免疫组化染色法,对43例食管癌手术标本的CyclinD1、CDK4蛋白进行标记。结果在鳞癌组织中,两种抗体的阳性物质存在于细胞核中和/或胞浆中,在正常鳞状上皮主要存在于核中,且从正常鳞状上皮→癌旁不典型增生→鳞癌组织,CyclinD1、CDK4的阳性率逐渐上升,统计学处理:正常上皮与癌组织之间的阳性率有差异显著性(PCyclinD1<0.05,PCDK4<0.05);但未检出CyclinD1、CDK4与癌分化的关系。另外,在正常鳞状上皮中,CyclinD1、CDK4则多集中在基底层的增殖带。结论CyclinD1、CDK4蛋白表达的变化是食管癌发生中的早期事件,并且在正常细胞周期中起正调控作用。Abstract In order to study the role of CyclinD 1、CDK 4 gene and protein in the carcinogesis of esophageal Cancer,as well as the interation between them.CyclinD 1、CDK 4 of the specimens from 43 cases of esophageal squamous cell carcinomas were labelled with immunohisto chemical LSAB method.The results are as fouows:The positive substance of CyclinD 1、CDK 4 were all seen in nucleus and cytoplasm in cancer tissue,they were mainly seen in nucleus in nomal epithelium;With the change from the nomal epithelium to displasis to squamous cancer tissue,the number of CyclinD + 1、CDK + 4 increased gradually.There were significant positive rate CyclinD 1、CDK 4 between the cancer tissue and the nomal epithelium (P CyclinD 1 <0 05,P CDK 4 <0 05).But there were not significant positive rate difference of CyclinD 1、CDK 4 among each grade.We also found higer positive expression rates of CyclinD 1 and CDK 4 in the spinous cells of the nomal epithelia.The Study domonstrated that the alteration of CyclinD 1、CDK 4 protein is an early event in the Carcinogenes of esophaged cancer,In addition,CyclinD 1、CDK 4 gene provide a positive control regulation for nomal cell cycle.
