摘 要:目的:研究核仁组成区嗜银蛋白定量分析技术与DNA流式细胞术在鼻腔癌及乳头状瘤诊断中的相关性。方法:用流式细胞术(FCM)及核仁组咸区嗜银蛋白(Ag-NORs)定量分析技术对鼻腔癌,乳头状瘤及正常鼻腔粘膜组织共50例石蜡切片标本细胞核DNA含量与AgNORs颗粒数的变化作定量分析和两者之间的相关性研究。结果:(1)DNA含量变化和AgNORs颗粒数都随病变程度的加重而增加,且良恶性肿瘤之间和恶性肿瘤不同组织病理学之间都有显著差异(P<0.01)。(2)AgNORs颗粒数量与DNA流式细胞术所测的DI、PI、s%之间都有明显的直线相关性(r=0.7333,0.7688,0.8721,P<0.001)。结论:AgNORs颗粒数与DNA含量的变化对鼻腔癌及乳头状瘤的诊断与研究具有相近的意义。AgNORs计数分析染色简便,设备便宜,易于推广。Objective: Study correlation between nucleoar organizer region data and DNA flow cytometry in nasal cancer and papilloma. Metheds: DNA content and silverbinding nucleolarorgarnzer region(AgNOR) data in 50 cases of paraffin-embedded nasal cancer, papillorma andnormal mucosa tissues were studied quantitatively using DNA flow cytometry and argytrophilic staining technique. The correlation between DNA content and AgNOR data was alsoanalyzed. Results: (1)DNA content and AGNAR content increased with the elevation of theseverity of the pathological changes and there were significant differences in them betweenbenign and malignant neoplasms and arnong neoplasms developed from different tissues(P<0. 01 ). (2)There was a very gdri linear correlation between the mean numbers of AgNORsites per nucleus and DI, PI, or s%, Obtained from DNA flow cytometry of the same tissue(r= 0. 7701, 0. 8619, 0. 8956, respectively P<0.001). Conclusion: AgNOR data and DNAcontent are with similar values in diagnosis and the further research of nasal cancer and papilloma. It is also proposed that as the staining and counting techniques used in assay of AgNOR is simple and inexpensive, they may thus be applied widely.
