老化红细胞变形能力与膜磷脂及收缩蛋白含量的关系

摘　　要：①目的探讨老化红细胞变形能力与红细胞膜磷脂成分和收缩蛋白（ＳＰ）含量的关系。②方法检测４２例健康人外周血老化和年轻红细胞滤过指数（ＥＦＩ），膜磷脂成分中的神经磷脂（ＳＭ）、磷脂酰胆碱（ＰＣ）、磷脂酰丝氨酸（ＰＳ）、磷脂酰乙醇胺（ＰＥ）、膜ＳＰ二聚体（ＳＰ－Ｄ）及四聚体（ＳＰ－Ｔ）含量的变化。③结果老化红细胞ＥＦＩ明显高于年轻红细胞（ｔ＝４．０６０，Ｐ＜０．００１），老化红细胞膜ＳＭ，ＳＭ／ＰＣ，ＳＰ－Ｄ和ＳＰ－Ｄ／ＳＰ－Ｔ明显增高，而ＰＥ，ＰＳ，ＰＣ和ＳＰ－Ｔ明显减低，与年轻红细胞比较差异有极显著性（ｔ＝２．９１１～３．５２０，Ｐ＜０．０１，０．００１）；老化红细胞ＥＦＩ与ＳＰ－Ｄ，ＳＰ－Ｄ／ＳＰ－Ｔ，ＳＭ和ＳＭ／ＰＣ呈正相关（ｒ＝０．５１１～０．６８４，Ｐ＜０．００１），而与ＳＰ－Ｔ，ＰＥ，ＰＳ，ＰＣ呈负相关（ｒ＝－０．５５２～－０．６１４，Ｐ＜０．００１）。④结论老化红细胞变形能力降低与膜磷脂成分和膜ＳＰ异常有密切关系。Objective To explore the the relationship between senescent erythrocyte deformability and lipid composition, spectrin content(SP) of erythrocyte membrane. Methods The erythrocyte filtration index(EFI), the contents of sphigomyeli(SM), phosphatidylcholin (PC), phosphatidylserine(PS) and phosphatidyltholamine(PE) of phospholipids, SP dimers(SP D) and sptetramers( SP T) were measured in senescent and young erythrocyte of 42 healthy subjects. Results EFI was higher in senescent erythrocyte than that in young erythocyte( t=4.060, P <0.001); SM, SM/PC, SP D/SP T were higher, PE, PS,PC and SP T were lower in senescent erythrocyte than those in young erythocyte( t=2.911—3.520, P <0.01,0.001); Senescent erythrocyte EFI was positively correlated with SP D, SP D/SP T, SM and SM/PC( r=0.511—0.684,P <0.001), and negatively correlated with SP T,PE,PS and PC( r=-0.552— -0.614, P< 0.001). Conclusion The decrease of senescent erythrocyte deformability was closely correlated with the abnormalities of lipid composition and spectrin content in erythrocyte membrane.
