摘  要:目的:观察重组人白细胞介素3(rhIL3)刺激后AML细胞对柔红霉素(DNR)敏感性的变化。方法:利用流式细胞仪(FCM)、细胞形态学、联苯胺染色,观察rhIL3对AML细胞增殖与分化的影响;用荧光分光光度法测定AML细胞内DNR浓度;用半固体培养法观察CFUAML产率。结果:①rhIL3对体外培养的AML细胞具有促增殖作用,能增强AML细胞对DNR的敏感性;②rhIL3或rhIL3联合DNR对AML细胞无明显诱导分化作用;③rhIL3不影响AML细胞对DNR的摄取与排泄平衡。结论:本实验为rhIL3与DNR联合应用。Purpose: To evaluate the effect and the mechanism of recombinant human interleukin-3(rhIL-3) on the sensitivity of acute myeloblast leukemia(AML) cells to daunomycin(DNR). Methods: The effects of rhIL-3 of the proliferation and differentiation on AML cells were observed through the flow cytometry(FCM), cyto-morphology classification and benziaine dyeing. The intracellular concentration of DNR in AML cells was measured through fluorospectrophotometry. The CFUAML cell existing rate was countered through semisolid cultivation. Results: rhIL-3 can induce AML cells into cytoproliferation cycle and enhance its sensitivity to DNR, but it had no obvious effect on inducing differentiation of AML cells, rhIL-3 didn't break the balance between AML cells absorbing and excreting DNR. Conclusion: The combined using of rhIL-3 and DNR established an experimental base for increasing the clinical therapeutical effectiveness to AML treating.
