TGFβ_1及TGFβ_1 mRNA在病毒性肝炎肝组织中的表达

摘　　要：为了解慢性病毒性肝炎肝组织中转化生长因子β_1(TGFβ_1)及TGFβ_1mRNA的表达情况及其与肝脏病理改变的关系,本文用免疫组织化学法检测了45例慢性病毒性肝炎患者肝脏TGFβ_1表达,并做其中21例TGFβ_1mRNA原位杂交。结果TGFβ_1肝组织表达阳性率为88.89％,主要为间质细胞、汇管区及纤维化发生活跃部位阳性,且随肝纤维化程度加重而表达增强(r=0.73,P<0.05);TGFβ_1mRNA表达阳性率为57.14％,不仅定位于单核细胞、窦细胞,某些肝细胞也有表达。结果表明TGFβ_1与肝纤维化密切相关,不仅间质细胞能够产生TGFβ_1促进肝纤维化,肝细胞本身可能也参与TGFβ_1分泌环路,在肝纤维化发病机制中起一定作用。To investigate the relationship between expression of TGFβ_1, and its mRNA in liver tissues of patients with viral hepatitis and degree of liver fibrosis, TGFβ_1, protein was detected in 45 liver tissues from the patients with chronic viral hepatitis by immunohistochemical method and TGFβ_1, mRNA was detected in 21 of the 45 specimens by in situ hybridization. TGFβ_1 protein was positive in 40 of the 45 patients and was mainly distributed in interstitial area, portal area and fibrotic area, The expression of TGFβ_1, protein was positively correlated with degree of liver fibrosis (r = 0. 73. P<0. 05). TGFβ_1 mRNA was positive in 12 of 21 patients. Positive stains were detected not only in sinusoidal cells and mononuclears, but also in some parenchymal cells. These results show that the expression of TGFβ_1 correlated closely with liver fibrosis of viral hepatitis. It is possible that the parenchymal cells in liver might participate in the fibrogenesis by producing TGFβ_1.
